The QuantiNova kit family represents our most advanced generation of real-time PCR and RT-PCR kits and perform equivalent or better than QuantiFast and Rotor-Gene kits. The QuantiNova kits will replace the QuantiFast and Rotor-Gene PCR kit product lines, both of which will be discontinued in production by June 30, 2021, and afterward, offered only until stocks last. How do I ensure the optimal performance of the successor kit?
As a general rule, please do follow the protocol of the successor kit. It has been optimized for the newly developed kit composition and provides various benefits like increased speed, yield and specificity. You will only benefit from these improvements when following the recommended protocol. Refrain from copy-pasting protocols from other or older kits.

What are my ordering conditions for the successor kit?

Does Taq polymerase of QuantiNova and QuantiFast/Rotor-Gene kits differ?
No, both kits contain the same Taq polymerase. However, the way it is kept inactive differs. The QuantiNova kits use a unique antibody-mediated hot-start mechanism whereas the QuantiFast/Rotor-Gene kits use a chemical hot-start mechanism.
How does hot-start procedure of QuantiNova kits compare to QuantiFast or Rotor-Gene kits?
In QuantiFast and Rotor-Gene kits, Taq polymerase hot-start is controlled by a chemical mechanism. While chemical inactivation provides a highly stringent hot-start control, the reactivation is relatively slow. The QuantiNova kits employ a novel and stringent antibody-mediated hot-start control that allows reactivation of Taq polymerase in 2–3 minutes. Additionally, the enzyme is protected by a QuantiNova Guard molecule ensuring extraordinary stringency of hot-start for utmost specificity and stability.
Does reverse transcriptase of QuantiNova and QuantiFast/Rotor-Gene kits differ?
Yes, they are different. The QuantiNova kit family features a newly developed, RNaseH-deficient, Moloney Murine Leukemia Virus Reverse Transcriptase (M-MLV RT) while QuantiFast/Rotor-Gene Reverse Transcriptase is RNase H+.
The QuantiNova RT kits come with further benefits. The QuantiNova RT-PCR Kits for 1-step RT-PCR feature a unique 2-phase hot-start procedure, that allows reaction setup at room temperature and prevents unwanted, premature enzyme activity during reaction setup. The QuantiNova Probe RT-PCR Kits additionally integrate genomic DNA reduction providing increased precision in mRNA quantification.
Will visual pipetting dyes of QuantiNova kits interfere with PCR results?
No, they won‘t. The tracking dyes allow visual check of manual or automated pipetting. They have been thoroughly tested, and have been in use for several years now. We have not experienced any difficulties.
Why is ROX dye provided in a separate tube in QuantiNova kits?
This offers highest flexibility if using ABI cyclers that require ROX as reference dye. A different amount of ROX is added before use to fit the requirements of “Low ROX” and “High ROX” cyclers, as clearly outlined in our handbook. For other cyclers that typically do not require ROX as reference dye, the respective channel could be used for target detection.
Why do QuantiNova RT-PCR and Reverse Transcription kits include Internal Control (IC) RNA?
IC RNA is an important safety measure to specifically monitor the presence of inhibitors. Its use is optional, and the respective SYBR Green- or probe-labelled IC assays are available as accessories in our (web) catalog. Inhibitors carried over from RNA extraction or cDNA synthesis will not necessarily lead to complete failure of the qPCR reaction, but can compromise PCR efficiency, shifting the CT or Cq values. This effect is neither visible nor can be quantified, potentially leading to significant, non-reproducible mis-quantification in real-time PCR results. Inclusion of an internal control RNA can avoid these effects and is a standard requirement in IVD real-time PCR procedures. We recommend its use also for real-time PCR in gene expression studies, as it is provided in a convenient and easy-to-use format in our QuantiNova kits.
Will I get different results or measured values when I switch from QuantiFast/Rotor-Gene kits to QuantiNova kits ?
All results should remain highly consistent and comparable between both kit families. However, single assays might behave slightly different and RFU values for QuantiNova SYBR Green PCR kits are higher than for QuantiFast/Rotor-Gene kits, to provide a more consistent and robust signal (this does not affect CT or Cq values.)
To ensure optimal results right from the start, we highly recommend to use the standard reaction setup protocol and cycling profiles outlined in the QuantiNova Quick Start Protocol and Handbook. This way you can fully benefit from the improved QuantiNova features.
Is the Rotor-Gene Q instrument affected by this reagent kit discontinuation?
No, the Rotor-Gene Q instrument is not affected. We will continue to offer it as an integral component of our assays technologies portfolio.
I’m using QuantiFast Pathogen (RT-) PCR + IC Kits (or QuantiFast Multiplex RT-PCR Kits) in validated procedures like detection of SARS-CoV-2 or other pathogens. Are these products affected by this phase-out?
No, QuantiFast Pathogen PCR + IC Kits (Cat. no. 211352, 211354), QuantiFast Pathogen RT-PCR + IC Kit (Cat. no. 211454) and Internal Control DNA/RNA (Cat. no. 211392, 211492) are not affected by this phase-out and will remain in our portfolio. The QuantiFast Multiplex RT-PCR +R Kit (2000) (Cat. no. 204956) will also remain available. However, smaller pack sizes of the QuantiFast Multiplex RT-PCR Kit will be offered only until June 30, 2021.
