原理

QIAamp One-For-All Nucleic Acid Kit使用快速真空操作,简化从多种类型样本中纯化病毒RNA和DNA的操作流程,无需苯酚-氯仿抽提。核酸特异性结合到QIAamp硅胶膜上,污染物流出。通过三步高效洗涤完全去除二价阳离子和蛋白质等PCR抑制剂,再用水或试剂盒中的缓冲液洗脱结合在离心柱上的纯DNA。

QIAamp样本制备技术完全得到验证认可。

程序

试剂盒提供优化的缓冲液高效裂解样本,稳定核酸并促进其选择性结合到QIAamp硅胶膜上。裂解液中添加乙醇并上样到QIAamp 96孔板。使用洗涤缓冲液去除杂质,洗脱的病毒核酸、基因组DNA和胞内RNA可即时用于扩增反应或–20ºC贮存。纯化的核酸不含蛋白质、核酸酶或其他杂质。

What can be used as an alternative to the A260 measurement for quantification of small amounts of RNA and DNA?

Small amounts of RNA and DNA may be difficult to measure spectrophotometrically. Fluorometric measurements, or quantitative RT-PCR and PCR are more sensitive and accurate methods to quantify low amounts of RNA or DNA.

Fluorometric measurements are carried out using nucleic acid binding dyes, such as RiboGreen® RNA Quantitation Reagent for RNA, and PicoGreen® DNA Quantitation Reagent for DNA (Molecular Probes, Inc.).
