artus HSV-1/2 PCR Kits是使用real-time PCR技术的即用型分子检测试剂盒。该试剂盒包含体外诊断中对脑脊液((RG和QS-RGQ试剂盒)、血浆、血清和拭子(LC和TM试剂盒)中纯化的HSV-1/2 DNA进行快速、灵敏检测的所有所需试剂,并已经过优化。artus HSV-1/2 LC、RG和TM PCR Kits使用自动或手动样本制备和手动反应体系构建技术。artus HSV-1/2 RG PCR Kit另有EASYartus HSV-1/2 RG PCR Kits,可与用于自动纯化病毒核酸的EZ1 DSP Virus Kit配合使用。artus HSV-1/2 QS-RGQ Kit可在QIAsymphony RGQ仪器上使用,从样本制备到病原体检测均为完全自动完成,不可进行手动反应体系构建。

原理

artus HSV-1/2 PCR Kits的原理为使用real-time PCR技术对HSV-1和HSV-2基因组特定片段进行扩增和实时检测。该试剂盒特异性强、灵敏度高(参见" Highly sensitive detection of HSV-1 DNA"),且重复性好。

每个试剂盒中都包含HSV-1和HSV-2的阳性对照(参见" Detection of the HSV-1 positive control"和" Detection of the HSV-2 positive control")。此外,该试剂盒还含有异源扩增系统,即内参,可识别PCR抑制剂。内参(IC)可在PCR中通过不同的荧光通道检测。

Should carrier DNA or RNA be used for nucleic acid extraction of Varicella Zoster Virus (VZV) and Herpes Simplex Virus (HSV) samples for artus® assays?

In general and especially for extraction from cerebrospinal fluid, serum or plasma, QIAGEN recommends the use of carrier RNA during nucleic acid extractions to increase the yield of extracted pathogen DNA. QIAGEN recommends to use Poly(A) RNA homopolymers from GE Healthcare (Cat. no. 27- 4110-02).

The artus HSV-1/2 RG PCR kit detects HSV-1/2 specific DNA. Other Herpes viruses are not detected. The LOD is 0.12 copies/µl for HSV-1 and 0.16 copies/µl for HSV-2

Does the artus HSV/2 RG PCR kits cross-react with any other pathogens?
No. The artus HSV-1/2 RG PCR Kit is highly specific and shows no cross-reactivities. Sample materials with the following pathogens tested negative: EBV, VZV, CMV, HHV6, HAV, HBV, HCV, HIV, WNV, Parvo B19, Enterovirus.

Why are the fluorescence intensities of the HSV detection markedly lower than those of the HSV-1 detection using the artus® HSV 1/2 LC PCR Kit?

The artus® HSV-1/2 LC PCR Kit contains probes for both the HSV-1 and HSV-2 detection. Yet, the hybridization properties of these probes differ with the template DNA (HSV-1 or -2) to allow for a melting curve based virus type discrimination. The probes which match HSV-1 perfectly carry a few mismatches for HSV-2 leading to decreased fluorescence signal.
