Product Details

The Investigator Argus Y-28 QS Kit allows multiplex amplification of 27 Y-chromosomal STR markers from multiple male-specific sample types, such as male–male and male–female mixtures, as well as direct PCR amplification from single-source samples. Due to the choice of markers, of which six are rapidly mutating (RM), the kit offers best-in-class discrimination power. In addition, the kit includes the innovative Quality Sensor, which generates additional, valuable data for your quality control and performance checks. The kit uses QIAGEN's latest fast-cycling technology FRM 3.0, which not only enables PCR in around 90 minutes, but also provides enhanced stability and inhibitor tolerance. Highly sensitive, yielding rapid and reliable results from trace DNA, the Investigator Argus Y-28 QS Kit is suitable for Y-STR casework, database samples and paternity testing.

Performance

The Investigator Argus Y-28 QS Kit uses our latest FRM 3.0 technology and provides reliable results in 90 minutes from casework samples or, in a direct amplification approach, from reference samples. For amplification from purified single source samples, we also offer a fast cycling protocol that takes only ~70 minutes. This proven QIAGEN multiplex PCR technology has been optimized for ease of handling and increased sensitivity. The Investigator Argus Y-28 QS Kit is optimized to improve the resolution of trace evidence and sexual assault mixtures that contain small quantities of male DNA in a high background of female DNA, and offers laboratories both powerful discrimination and high performance to enable accurate results from challenging samples. Assay performance tests in the presence of increasing concentrations of inhibitors showed both high sensitivity and superior inhibitor resistance. This means reliable, high-quality results.
The Investigator Argus Y-28 QS Kit was validated and evaluated using the Veriti and GeneAmp 9700 thermal cyclers and the Applied Biosystems 3500 Genetic Analyzer. The recommended amount of DNA under standard conditions is 0.5 ng. Internal validations demonstrated robust and balanced results with 0.1 to 2 ng DNA, and reliable results with <0.1 ng DNA.

Principle

The Investigator Argus Y-28 QS Kit uses QIAGEN's fast-cycling PCR technology for the rapid, simultaneous amplification of 27 Y-chromosomal STR markers and the Quality Sensor. Carefully designed primers minimize the overlap of alleles of different makers, thereby minimizing the likelihood of data misinterpretation.
The innovative Quality Sensor provides additional information about the sample quality and PCR success beyond the profile of the sample. With two Quality Sensors amplified as a positive control, it is possible to determine:
• If the PCR amplification worked
• If there was any DNA in the sample
• If the sample was degraded
• If inhibitors in the sample prevented successful PCR
This additional information will greatly enhance how you analyze your data and aid you in making crucial decisions with respect to further sample processing. Ultimately, the Quality Sensor will help you reduce unnecessary re-runs, saving you time and money.

Procedure

The 27 Y-chromosmal STR markers, and the Quality Sensor, are amplified simultaneously in a single PCR and labeled with one of the dyes listed below in Table 1.

The amplified loci then undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as Applied Biosystems GeneMapper ID-X. See the kit technical specifications for more information.
