Product Details

The QIAgility is a compact benchtop instrument that enables rapid, high-precision setup of PCR experiments with ready-to-use software for ease and convenience. The software provides step-by-step guidance for worktable setup and automatic calculation of all mixes, eliminating the need to program pipetting steps. Just select the kit and cycler, define your targets and start the run. For added flexibility the QIAgility is operated via a laptop computer.

Unmatched pipetting precision with QIAgility

The QIAgility instrument operated by the QIAgility Setup Manager Software demonstrates exceedingly high accuracy and precision of dispensed volumes as verified by the Artel MVS system (see figure “ Exceptional pipetting precision and accuracy spanning over a broad volume range”). These results indicate that the QIAgility can produce automated, high-performance liquid dispensing results, while avoiding common errors that can be associated with manual pipetting and thereby reducing the time to result.

Regardless of whether you pipet water or QIAGEN ready-made reaction mix to set up a PCR experiment, the QIAgility instrument can dispense volumes that are highly precise and also generate significantly reproducible replicates (see figure “ Unrivaled pipetting precision and reproducibility of the QIAgility for master mix solutions”).

High performance PCR setup using QIAgility

A QuantiNova SYBR Green PCR assay setup was performed manually and automated in parallel using the QIAgility along with its Setup Manager Software. The amplification run was done on the Rotor-Gene Q and resulting data analyzed using the Q-Rex Software. Consistent results were obtained from both manual and automated reaction setup using the same template dilutions (see figure “ Comparison of manual and automated assay setup using series dilutions”).

Exceptional linearity of QIAgility

QIAgility exhibits excellent linearity and assures accurate and reliable use of sample for an assay. To assess the linearity of the system, pure gDNA was serially diluted using the QIAgility and measured for their concentrations on the QIAxpert. The quantifications from the QIAxpert were plotted against the known concentration values, and the resulting plot demonstrates the exceptional linearity (with a regression coefficient of >0.999) of QIAgility (see figure “ High system linearity of QIAgility”).

Principle

Automated PCR setup is rapid and reliable and eliminates manual pipetting steps that are typically prone to human error.

Liquid-handling tasks are performed by the single-channel pipetting head, which provides fast transfer of liquids with various types of plasticware. For example, 96 samples with the QuantiFast SYBR Green PCR Kit with 2 standard series can be setup in approximately 45 minutes. The worktable features a flexible layout and supports almost all kinds of cyclers and labware, giving high application versatility. Dynamic liquid-level sensing, facilitated by conductive filter tips offers maximum safety control.

The advanced design of the QIAgility delivers improved process safety, giving you peace of mind. A built-in sensor ensures that the instrument pauses when the hood is opened during a run, protecting users from moving parts. The UV light ensures effective worktable decontamination and the HEPA filter provides positive clean air pressure throughout PCR setup.

QIAgility is a compact benchtop instrument with a very small footprint. It fits easily onto most lab benches and can be readily moved into biosafety level laboratories. For added flexibility, the QIAgility comes with a laptop computer. To reduce your work load, comprehensive run reports containing all run information are generated automatically during each protocol.

This study tested a workflow for quantitation and qualification of AAV samples using a duplex assay on the QIAcuity dPCR instrument targeting both an insert (GFP) and the viral backbone (AAV2-ITR). With very low intra-assay and inter-assay CVs <6.5%, we demonstrate one of the main benefits of dPCR: reproducibility.
The QIAgility instrument is a liquid handler designed for automating PCR setup. For compatibility with the QIAcuity, we developed an adapter to secure up to two nanoplates onto the deck of the QIAgility. Using the QIAgility software, we have optimized a protocol that works for all nanoplate types and QIAcuity applications. Here we report the performance of a front end automated QIAgility dPCR nanoplate setup procedure for use with the QIAcuity dPCR system.

FAQ

What are the advantages and disadvantages of turning on the tip “re-use” feature on the QIAgility?

Pros: saves tips and therefore cost.

Cons: lowers accuracy and precision as liquid is built up on the inner and outer walls of the tip with each use.

How can I decontaminate my QIAsymphony SP / AS system?

The QIAsymphony system has an integrated UV lamp. UV decontamination should be performed daily. It helps to reduce possible pathogen contamination of the QIAsymphony SP/AS worktables. The efficiency of inactivation has to be determined for each specific organism and depends, for example, on layer thickness and sample type. QIAGEN cannot guarantee complete eradication of specific pathogens.

How do I transfer the QIAgility calibration settings to a different computer?

The QIAgility calibration settings can be saved using a “Setup Disk” and transferred to a different computer. This is useful and time-saving when using a different computer to run the instrument. Calibration on the new computer is not required if the calibration settings are transferred.

 

The instructions for creating and loading calibration settings from a “Setup Disk” are outlined in the QIAgility User Manual section 5.5.1.

 

Note: A “Setup Disk” created for one instrument should never be used with another instrument, as this may result in damage to the instrument.

What file is required for reporting QIAgility issue?

The Support Packages are important when reporting an issue to QIAGEN Technical Services. Upon completion of each run, the software automatically saves a support package.

 

To retrieve a support package, click Help in the QIAgility software and select Choose support package. In the new window, select the support package that corresponds to the issue and send it to QIAGEN Technical Service.

What causes Error 131?

A malfunctioning HEPA unit is the most common cause of Error 131. Shut down the system and disconnect the HEPA filter unit from the QIAgility instrument. Power on the instrument and launch the QIAgility software. If the error goes away, the HEPA filter unit needs replacing.

Ensure that correct volumes of reagent are added to the tubes. Re-perform the tube height calibration.

What do I do if a red exclamation icon appears in the top-right corner of the schematic plate on the main software window?

The red exclamation icon appears if the plate has not been position- or height-calibrated, and will remain visible until all of the calibrations for that plate have been completed. Perform the position and height calibration as outlined in the QIAgility User Manual.

What do I do if the pipettor hits the tip rack?

Ensure the tip rack is correctly oriented and pushed towards the upper-left corner of the deck slot. Perform the tip rack position calibration so that the pipettor hub is in the center of an empty tip position hole. Do this for all tip racks on the deck.

Vapor-Lock is fully compatible with the QIAgility instrument for high-precision, automated reaction setup. What sample tubes can I use on my QIAsymphony SP system?

Please navigate to the Product Page of the QIAsymphony Kit you are using; for example, the QIAsymphony RNA Kit, Cat No./ID: 931636.  Refer to the resources tab and download the labware document. The labware document will list the sample tubes that have been tested for your application. Please contact Technical Service if you wish to use sample tubes not listed in the labware document.

Why do I do if the QIAgility software runs in virtual mode while the instrument is connected to the computer?

Make sure the QIAgility software uses the same COM port number as assigned to the QIAgility instrument on the computer. The COM port setting can be checked by clicking Options\Robot Setup\Select COM port in the QIAgility software.

 

Ensure that the cable between the QIAgility instrument and the computer is securely inserted. Power on the instrument first, and then launch the QIAgility software. 

 

The QIAgility has both RS-232 (serial) and USB interfaces for computer connection. Try an RS-232 cable if the communication cannot be established between the instrument and computer with a USB cable.

What do I do if tips are not ejected?

Tip ejector calibration must be performed before any other calibration. Perform tip ejector calibration as outlined in the QIAgility User Manual.

 

During calibration of the tip ejector, the pipettor hub should be centered inside the slot of the tip ejector in the Y-axis — it should not touch either side.
