The QIAamp UCP Pathogen Mini Kit is for microbial DNA purification from small volumes of whole blood, swabs, cultures, and body fluids.

 

Performance

The QIAamp UCP Pathogen Mini Kit, when used with Pathogen Lysis Tubes, provides efficient cell lysis, with high yields of pathogen DNA. Pathogen Lysis Tubes with bead types S or L are available as accessories.

The QIAamp UCP Pathogen Mini Kit purifies microbial DNA for use in downstream applications. Ultraclean production (UCP) reagents and consumables provide high-quality purified DNA for your downstream applications. The QIAamp UCP Mini Columns and all buffers within the QIAamp UCP Pathogen Mini Kit undergo DNA decontamination processes and are certified for the functional absence of bacterial and fungal DNA at the time of delivery.

 

Principle

The QIAamp UCP Pathogen Mini Kit enables purification of microbial DNA using tailored sample pretreatement protocols with simplified sample prep protocols (spin protocol or vacuum protocol). Pretreatment protocols include mechanical disruption using Pathogen Lysis tubes (available as accessories) for maximal lysis efficiency for cells of gram-positive bacteria, yeast, and other fungi. Using the QIAvac 24 Plus with the QIAamp UCP Pathogen Mini Kit enables starting samples of up to 400 µl and flexible elution volumes from 50 to 200 µl. Multiple samples may be processed simultaneously, providing pure nucleic acids in less than 2 hours per 24 samples.

 

Procedure

The QIAamp UCP Pathogen Mini Kit uses tailored sample pretreatment protocols with sample prep protocols. Using Pathogen Lysis Tubes for mechanical disruption provides efficient lysis for cells of Gram-positive or Gram-negative bacteria, as well as yeast, and other fungi.

What can be used as an alternative to the A260 measurement for quantification of small amounts of RNA and DNA?

Small amounts of RNA and DNA may be difficult to measure spectrophotometrically. Fluorometric measurements, or quantitative RT-PCR and PCR are more sensitive and accurate methods to quantify low amounts of RNA or DNA.

Fluorometric measurements are carried out using nucleic acid binding dyes, such as RiboGreen® RNA Quantitation Reagent for RNA, and PicoGreen® DNA Quantitation Reagent for DNA (Molecular Probes, Inc.).
