What do I do if I cannot see amplification in the FAM channel in my KRAS and EGFR PCR kits?

If there is no amplification in the FAM channel, check for exogenous control signal. The exogenous control reaction is included in all master mixes. It should be positive in all reactions. If it is positive but the control assay is negative for the FAM reaction, this suggests there is little or no amplifiable DNA in the reaction. If the control assay does give a FAM signal but the mutation reactions are negative, providing there is an exogenous control signal, the sample is negative for the mutations.
