Product Details

The Investigator HDplex Kit amplifies 12 autosomal STR markers, most of which are not featured in commonly used marker standards. This enables the reliable differentiation of forensic samples derived from related individuals and gives heightened discriminatory power in these demanding cases. The genetic analysis is calibrated with BT5 Matrix.

The Investigator Triplex AFS QS Kit is for forensic stain pre-selection, which serves as a starting point for genotyping strategies. It enables simultaneous amplification of STR loci FGA (FIBRA), SE33 (ACTBP2) and Amelogenin from forensic or reference samples. The genetic analysis is calibrated with the DS-30 matrix.

Investigator STR Kits meet ISO 18385 requirements.

Performance

Investigator HDplex and Triplex AFS QS Kits have been validated using the GeneAmp 9700 thermal cycler and the Applied Biosystems 3130 Genetic Analyzer.

The recommended amount of DNA under standard conditions is 0.35 ng. Internal validations using the Investigator HDplex Kit demonstrated robust and balanced results with 0.2–1.0 ng DNA, and reliable results with <0.1 ng DNA. Internal validations using the Investigator Triplex AFS QS Kit demonstrated robust and balanced results with 0.1–1.0 ng DNA, and reliable results with <0.1 ng DNA.

Principle

The Investigator HDplex Kit enables the simultaneous amplification of 12 STR markers (D2S1360, D3S1744, D4S2366, D5S2500, D6S474, D7S1517, D8S1132, D10S2325, D12S391, D18S51, D21S2055, SE33) and the gender-specific Amelogenin. These markers include some highly polymorphic STR markers that are not included in commonly used standards enabling higher discrimination in difficult cases.

The Investigator Triplex AFS QS Kit enables the simultaneous amplification of the two polymorphic STR loci FGA (FIBRA) and SE33 (ACTBP2), and the gender-specific Amelogenin. The kit also includes the Quality Sensor (QS). This special feature is an internal PCR control that provides information on the presence of PCR inhibitors and the efficiency of the PCR.

Procedure

With the Investigator HDplex Kit, the 12 polymorphic STR markers and Amelogenin are simultaneously amplified and labeled with the dyes listed in the table "Investigator HDplex Kit dyes and corresponding markers".

Using the Investigator Triplex AFS QS Kit, FGA (FIBRA), SE33 (ACTBP2) and Amelogenin are simultaneously amplified in a single PCR and labeled with 6-FAM.

The amplified loci then undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as Applied Biosystem’s GeneMapper ID-X Software. See the table "Technical specifications" for more information.

Applications

The Investigator HDplex Kit enables reliable generation of DNA profiles from blood, buccal swabs and forensic stains. Thanks to the STR markers used, this kit is highly suited for difficult forensic cases involving related individuals, difficult paternity cases, population genetic studies and other human identity testing applications involving related individuals.

The Investigator Triplex AFS QS Kit is preferentially used for forensic prescreening strategies or for low-copy and degraded DNA samples that do not give distinct results with complex multiplexes. It enables rapid and reliable DNA genotyping of blood, buccal swabs and forensic stains.
