The QIAcuityDx Universal MasterMix Kit contains a 4x concentrated, ready-to-use dPCR master mix optimized for microfluidic use with QIAcuityDx Nanoplates. The kit enhances dPCR specificity and efficiency for accurate quantification of DNA or cDNA targets with TaqMan probes. Reactions can be set up as either singleplex or multiplex, for up to 5-plex dPCR.
The QIAcuityDx Universal MasterMix Kit is used together with the QIAcuityDx System and QIAcuityDx Nanoplates for in vitro diagnostic applications.

Consistent performance
The QIAcuityDx Universal MasterMix contains our latest-generation, high-quality QuantiNova DNA Polymerase. This, in combination with our unique proprietary and proven buffer technology, delivers highly consistent results in terms of sensitivity, reproducibility and efficiency.
Probe-based detection from singleplex to 5-plex with the QIAcuityDx Universal MasterMix
The special master mix in the QIAcuityDx Universal MasterMix Kit enables accurate quantification of up to 5 targets in a well of the QIAcuityDx Nanoplate. This saves time and money and reduces the amount of sample material needed. Moreover, the duplex or multiplex dPCR data obtained are comparable with those obtained from a singleplex dPCR.
In-use stability
Once opened, reagents can be stored in their original packaging at –30 to –15°C until the stated expiry date shown on the packaging. Repeated thawing and freezing should be avoided. Do not exceed a maximum of five freeze–thaw cycles.
The reagents must be fully thawed at room temperature (15–25°C) for maximum of 30 minutes before use.
The QIAcuityDx Universal MasterMix Kit comprises a ready-to-use dPCR master mix containing reaction chemistry in PCR Buffer and proprietary reference dye, and separate tubes of 200mM magnesium chloride (MgCl2) 100% w/w and RNase-free water 100% w/w. A full list of materials to be used with the QIAcuityDx Universal MasterMix Kit can be found in the QIAcuityDx System User Manual.
The protocol is optimized for the quantification of DNA or cDNA targets using the QIAcuityDx Universal MasterMix Kit with TaqMan probes in a singleplex or multiplex reaction using the QIAcuityDx System.
The principle of the dPCR reaction in the nanoplates is described here.
Just like in a qPCR workflow, sample preparation includes the transfer of a final sample mix (comprised of master mix, primers/probes and sample) to the QIAcuityDx Nanoplate. The system integrates partitioning, thermocycling and imaging into a single, semi-automated instrument that achieves sample to result in approximately 2 hours. Analysis is performed using the QIAcuityDx Software Suite which can be connected to the QIAcuityDx Four via a direct Ethernet connection, or via the same local area network (LAN). This provides the concentration in copies per microliter of your target sequence and can also be used for quality control, such as for positive samples or No Template Control (NTC).
The QIAcuityDx Universal MasterMix Kit, in combination with the QIAcuityDx System enables quantitative analysis of DNA and cDNA targets for use in applications, including:
