Product Details

The QIAamp DNA Blood BioRobot MDx Kit provides automated DNA purification on the BioRobot MDx workstation or the BioRobot Universal System using proven QIAamp silica-membrane technology. The fully automated procedure on the BioRobot MDx workstation requires less than 2.5 hours, including bar code reading and complete process documentation, with no hands-on time during the run.

Performance

The QIAamp DNA Blood BioRobot MDx Kit process provides DNA isolation on a 96-well plate, automating processing of 32 to 96 samples (200 µl sample volume) in increments of 8 on the BioRobot MDx or the BioRobot Universal System.  The fully automated procedure, including bar code reading, 20-minute load check, and complete process documentation, requires less than 2.5 hours for 96 samples, with no hands-on time. The typical yield is 3–6 µg per 200 µl healthy whole blood, with an elution volume of 200 µl.

The resulting DNA performs well in downstream amplification and Southern blotting assays (see figures " No PCR inhibition" and " Reliable real-time PCR").

Principle

The QIAamp DNA Blood BioRobot MDx Kit simplifies isolation of DNA from whole blood with a fast, automated 96-well–plate procedure. No phenol–chloroform extraction is required. DNA binds specifically to the QIAamp silica-gel membrane while contaminants pass through. PCR inhibitors, such as divalent cations and proteins, are completely removed in three efficient wash steps, leaving pure DNA to be eluted in either water or a buffer provided with the kit. QIAamp technology yields genomic and mitochondrial DNA from whole blood ready to use in PCR and blotting procedures.

The advantages of the fully automated system include high process reliability and walkaway automation, to provide a highly efficient system for DNA purification for laboratories. QIAamp sample preparation technology is fully licensed.

Procedure

Optimized buffers lyse samples, stabilize nucleic acids, and enhance selective nucleic acid adsorption to the QIAamp membrane. Alcohol is added and lysates loaded onto the QIAamp 96-well plate. Wash buffers and ethanol are used to remove impurities, and pure, ready-to-use DNA is then eluted in water or low-salt buffer (see flowchart " protocol").

The kit can process samples from whole blood or buffy coat with common anticoagulants such as citrate and EDTA. The QIAamp DNA Blood BioRobot MDx Kit can process blood and buffy coat samples that are fresh or frozen, provided they have not been frozen and thawed more than once. QIAamp technology yields genomic and mitochondrial DNA from whole blood that is ready to use in PCR and blotting procedures.

What is the difference between QIAGEN Protease and QIAGEN Proteinase K provided in various QIAamp Kits?

QIAGEN Proteinase K is a subtilisin-type protease, which cleaves at the carboxyl side of hydrophobic, aliphatic and aromatic amino acids. It is particularly suitable for short digestion times. It possesses a high specific activity over a wide range of temperatures and pH values with substantially increased activity at higher temperature. Soluble calcium is not essential for enzymatic activity. This means that EDTA, which is used to inhibit Mg2+-dependent enzymes such as nucleases, will not inhibit Proteinase K activity.

QIAGEN Protease is a broad-specificity Serine protease with high activity, cleaving preferentially at neutral and acidic residues. It is an economical alternative to Proteinase K for isolation of native DNA and RNA from a variety of samples.

What can be used as an alternative to the A260 measurement for quantification of small amounts of RNA and DNA?

Small amounts of RNA and DNA may be difficult to measure spectrophotometrically. Fluorometric measurements, or quantitative RT-PCR and PCR are more sensitive and accurate methods to quantify low amounts of RNA or DNA.

Fluorometric measurements are carried out using nucleic acid binding dyes, such as RiboGreen® RNA Quantitation Reagent for RNA, and PicoGreen® DNA Quantitation Reagent for DNA (Molecular Probes, Inc.).

How do I safely inactivate biohazardous flow-through material?

Always dispose of potentially biohazardous solutions according to your institution’s waste-disposal guidelines. Although the lysis and binding buffers in QIAamp, DNeasy, and RNeasy kits contain chaotropic agents that can inactivate some biohazardous material, local regulations dictate the proper way to dispose of biohazards. DO NOT add bleach or acidic solutions directly to the sample-preparation waste. Guanidine hydrochloride in the sample-preparation waste can form highly reactive compounds when combined with bleach.
