How much DNA is needed in the PyroMark Q48?

We recommend to use between 0.5 and 2 pmol of template, which is the same amount needed for the other pyrosequencing platforms. For PCR, we recommend 10 ng DNA in 5 µl in a total PyroMark PCR reaction setup of 25 µl. (More DNA amount may be needed in case the DNA is fragmented, e.g. FFPE DNA). For Pyrosequencing, 10 µl of the PCR reaction should be used. The optimal template amount can of course differ between assays.
