BioSprint DNA Blood Kits provide all reagents and plasticware necessary for automated purification of genomic DNA from cultured cells, tissues, blood, dried blood spots and buccal swabs using proven magnetic-particle technology on the BioSprint 15 or BioSprint 96.

Purified DNA is up to 50 kb in size (see figures " Purification of high-quality genomic DNA from mouse tail" and " Reproducible purification of DNA from sheep ear tissue"). The DNA is ready to use in all downstream applications, including Southern blotting and enzyme applications such as PCR (see figure " High PCR performance of purified DNA").

Principle

BioSprint DNA Blood Kits are used with the BioSprint 15 or BioSprint 96. The kits use magnetic-particle technology for rapid DNA purification from cells, tissue and blood or blood-related products. BioSprint instruments control an array of magnetic rods that can attract or release magnetic particles and transfer them from tube to tube or well to well. The sequential transfer of magnetic particles allows a rapid purification procedure to be performed, from the initial binding of DNA to the particles, through to washing of the particles and elution of pure DNA (see figure " Efficient processing of magnetic particles"). Since the instruments transfer magnetic particles instead of liquids, they use minimal amounts of reagents, enabling cost-efficient sample preparation.

Procedure

Genomic DNA can be purified from cells, tissue and blood. Lysed samples are loaded into the instrument, which then performs all steps until pure DNA is obtained (see figure " BioSprint DNA procedure"). The kits provide all the necessary reagents and plasticware for sample preparation, and are available in different sizes, allowing cost-efficient use of consumables.

For purification of genomic DNA from tissue, dried blood spots or buccal swabs, Buffer ATL and QIAGEN Proteinase K are required (not supplied with BioSprint DNA Blood Kits).

Can the BioSprint instruments be placed in a cool room?

No, the BioSprint Instruments should not be operated in a cool room. Optimal operation temperature for BioSprint is 10-40°C. Operation at 4°C may harm the instruments.

Do you have stability data for genomic DNA isolated with the BioSprint Kits?

Yes. We have shown that DNA purified with the BioSprint DNA Kits (e.g., the BioSprint 15 DNA Blood Kit, BioSprint 15 DNA Plant Kit, BioSprint 96 DNA Blood Kit and BioSprint 96 DNA Plant Kit) is stable for at least 2 years when being stored at either 2-8°C or -20°C. Our data indicate however that DNA stability is dependent upon storage conditions. For additional information see also FAQ 518.

How do I perform a DNA precipitation to concentrate my sample?
  • Add 1/10 volume of 3 M Na-Acetate pH 5.2, and 2 to 2.5 volumes of ice-cold 100% ethanol to the DNA sample
  • Mix, and store at –20°C for at least 1 h to precipitate the DNA
  • Recover the precipitated DNA by centrifugation at full speed in a microcentrifuge for 15–20 min
  • Pour off the ethanol and wash the pellet twice with room-temperature 70% ethanol
  • Allow the DNA pellet to air-dry
  • resuspend the DNA in a suitable volume of sterile TE buffer or distilled water

Is magnetic particle carry-over in the eluates on the BioSprint 96 normal?

The BioSprint 96 instrument has a magnetic-particle collecting efficiency above 95%. However, to avoid particle carryover from the eluate plate to subsequent applications, you can use a plate magnet while pipetting.

What is the run time for the BioSprint 96 DNA Blood protocol on the BioSprint 96 instrument?

High-quality genomic DNA can be purified from up to 96 pre-lysed samples in just 20 minutes. Including setup, the total time per run for the blood and buffy coat protocol is 45 minutes. For the other protocols (e.g. tissue), the run time will be slightly higher depending on the time of pre-treatment.

How to quantitate DNA purified using the BioSprint 96?

Measure the absorbance of the eluate obtained on the BioSprint 96 instruments at 260 nm, 280 nm and 320 nm. Carryover of magnetic particles in the eluates may affect the A260 and A280 readings, but should not affect the performance of the DNA in downstream applications. Subtract the absorbance reading obtained at 320 nm from the readings obtained at 260 nm and 280 nm to correct for the presence of residual magnetic particles.

Does the Epitect Bisulfite Kit also work on DNA extracted from plants?

Bisulfite conversion of unmethylated cytosines into uracils with the EpiTect Bisulfite Kit works on DNA irrespective of the source organism. The DNA template needs to be of high purity for efficient conversion. We recommend to use genomic DNA extracted with our DNA isolation kits for clinical or animal and plant samples as a template for the EpiTect Bisulfite Kit.

How do I safely inactivate biohazardous flow-through material?

Always dispose of potentially biohazardous solutions according to your institution’s waste-disposal guidelines. Although the lysis and binding buffers in QIAamp, DNeasy, and RNeasy kits contain chaotropic agents that can inactivate some biohazardous material, local regulations dictate the proper way to dispose of biohazards. DO NOT add bleach or acidic solutions directly to the sample-preparation waste. Guanidine hydrochloride in the sample-preparation waste can form highly reactive compounds when combined with bleach.

Yes, the BioSprint 15/96 instruments are stand-alone instruments and do not need to be connected to a computer for the actual run. However, a computer is needed to import and export protocols.
