Product Details

The ipsogen JAK2 MutaQuant Kit is an in vitro molecular diagnostic kit for real-time PCR on the Rotor-Gene Q and other real-time PCR instruments. The kit provides reagents optimized for reliable and sensitive detection and quantification of the JAK2 V617F/G1849T mutation in genomic DNA extracted from peripheral blood of subjects with suspected myeloproliferative neoplasm (MPN).

Performance

To ensure highest sensitivity, the ipsogen JAK2 MutaQuant Kit has been optimized to detect the JAK2 V617F/G1849T mutation and uses prediluted plasmid standards and primers and probe mixes (see figures  Accurate detection of JAK2 V617F plasmid standards and  Reliable detection of JAK2 wild-type plasmid standards). The sensitivity has a limit of detection (LOD) of 0.061%. This optimal sensitivity can be obtained on specimens containing at least 10,000 copies of the JAK2 gene (wild-type and V617F mutation).

Principle

The ipsogen JAK2 MutaQuant Kits is a ready-to-use kit for the detection and quantification of the JAK2 V617F/G1849T somatic mutation due to a single nucleotide polymorphism (SNP) in genomic DNA extracted from peripheral blood of subjects with suspected myeloproliferative neoplasm (MPN) (see figure  JAK2 mutation site identification). The kit provides high levels of sensitivity and reproducibility. The ipsogen JAK2 MutaQuant Kit provides 4 plasmid standard dilutions for mutant JAK2 V617F and 4 for the wild-type JAK2 allele. Use of the standards enables accurate quantification of transcripts. Results are expressed as a percent of JAK2 V617F in total JAK2.

See figures

Procedure

The first step is extraction and determination of quality and quantity of genomic DNA. The second step is the amplification of DNA by real-time PCR. Using the ipsogen JAK2 MutaQuant Kit allows detection and quantification of the JAK2 V617F/G1849T mutation. Simply start the reaction using the optimized protocols described in the kit handbook.

Applications

The ipsogen JAK2 MutaQuant Kit enables sensitive and reliable detection and quantification of the JAK2 V617F/G1849T mutation for in vitro diagnostic use.
