Product Details

The EZ1 DSP DNA Blood Kit contains all required reagents and labware for automated purification of DNA from 200 or 350 µl human whole blood using magnetic particle technology. Reagents are supplied in prefilled reagent cartridges, which ensures speed and convenience in loading the EZ1 Advanced, the the EZ1 Advanced XL. or the EZ2 Connect MDx . The CE-IVD-marked system provides exact performance specifications, assuring highly reliable DNA purification.

Efficient purification of genomic DNA

Automated DNA purification provides high levels of reliability and standardization and helps to eliminate variation caused by handling errors. The EZ1 Advanced XL or the EZ2 Connect MDx in combination with the EZ1 DSP DNA Blood Kit enables efficient purification of genomic DNA from human whole blood.

 

Principle

Automated sample preparation enables efficient, standardized purification of nucleic acids. The EZ2 Connect MDx and the EZ1 Advanced XL enable purification of nucleic acids using proven magnetic-particle technology for use in routine in vitro diagnostic applications. Reagent production is subjected to stringent quality control assuring reliable and robust performance. An easy-to-use instrument, prefilled and sealed reagent cartridges, and simple protocol selection and worktable setup help to prevent handling errors and allow the system to be operated by anyone — from the novice to the expert. Manual handling of potentially infectious samples is minimized, ensuring safety to the user and reliable processing of samples. Purified genomic DNA is ready to use in sensitive downstream diagnostic assays, such as PCR.

Procedure

The EZ2 Connect MDx or the EZ1 Advanced XL in combination with the proven EZ1 DSP DNA Blood Kit enables purification of DNA from 1–24 samples (EZ2 Connect MDx) or 1-14 samples (EZ1 Advanced XL) of human whole blood. All processing steps are performed by the instrument, from piercing the reagent cartridge to elution of highly pure nucleic acids. The innovative design of the instrument means that the pipet tips function as separation chambers, increasing the efficiency of magnetic purification and eliminating the need for centrifugation steps. The streamlined procedure provides robust walkaway purification of nucleic acids with minimal hands-on time.

Applications

With the EZ2 Connect MDx or the combination of the EZ1 Advanced XL DSP DNA Blood Card and the EZ1 Advanced XL, the EZ1 DSP DNA Blood Kit provides a fully automated procedure for purification of DNA from human whole blood. Genomic DNA purified using the EZ1 DSP DNA Blood Kit is ready to use in downstream diagnostic assays.

How long can eluates be stored after sample preparation using the EZ1 instruments or EZ2 Connect MDx and the EZ1 DSP DNA Blood Kit?
We recommend storing the purified DNA at 2–8°C or at –20°C for up to 24 months. For extended storage time, we recommend storage at –20°C or –80°C for up to 36 months. The impact of DNA stability might be different for the specific downstream application being used and needs to be self-validated by the user.

What do I have to do if precipitates are visible in the lysis buffer (well 1 of the reagent cartridge [RCB])?
Place the reagent cartridges (RCB) into a shaker–incubator, and incubate at 30–40°C with mild agitation for up to 2 hours. Do not use the reagent cartridges (RCB) if the precipitates do not redissolve.

Can samples other than whole blood be used with the EZ1 DSP DNA Blood Kit?
No. The EZ1 DSP DNA Blood system is a generic system that uses magnetic-particle technology for automated isolation and purification of genomic DNA from human whole blood samples, for in vitro diagnostic purposes. The system is not for use with tissue or stool samples. It is also not for isolation or purification of bacterial, fungal, viral, or parasite nucleic acids, nor for isolation or purification of RNA. The system performance has not been evaluated for isolation or purification of genomic DNA from buffy coat, cultured or isolated cells, swabs, dried blood spots, or cell-free body fluids, such as liquor, plasma, serum, or urine. Contact your local QIAGEN representative for information about other EZ1 Kits.

How long can blood samples be stored before sample preparation using the EZ1 DSP DNA Blood system?
For short-term storage (up to 7 days), collect blood in tubes containing EDTA as an anticoagulant and store the tubes at 2–8°C. However, for applications requiring maximum fragment size, such as southern blotting, we recommend storage at 2–8°C for up to 3 days only, as low levels of DNA degradation will occur after this time. 

For long-term storage, collect blood in tubes containing a standard anticoagulant (preferably EDTA, if high-molecular-weight DNA is required) and store the tubes at −20°C for a maximum of 4 weeks. Longer storage may be possible depending on the downstream application but needs to be validated by the user.

Yield and quality of the purified DNA may depend on storage conditions of the blood. Fresh blood samples may yield better results. Do not re-freeze blood samples more than 2 times as this may result in decreasing DNA yield.

No. The EZ1 instruments or the EZ2 Connect MDx are intended to be used only with QIAGEN sample preparation cartridges.

How can I track samples during collection, sample preparation, and downstream assay?
A sample sheet template is included in the handbook. This sample sheet may be useful for recordkeeping when using the EZ1 DSP DNA Blood or the EZ1 DSP Virus procedure. This sheet can be photocopied and labeled with descriptions of the samples and details of the run.
