Product Details

The EZ1&2 RNA Tissue Mini Kit contains all required reagents and labware for automated purification of total RNA from up to 10 mg standard tissue samples using magnetic particle technology. Reagents are supplied in prefilled and sealed reagent cartridges, which ensures speed and convenience in loading the instrument.

The EZ1&2 RNA Tissue Mini Kit is automated with the EZ2 Connect (1–24 samples per run). This kit can also be used with the EZ1 Advanced XL (1–14 samples per run).

The yields and high quality of total RNA purified using the EZ1&2 RNA Tissue Mini Kit are consistent and reproducible (see figures " Efficient purification and reproducible yields" and " Consistent highly pure RNA").

Principle

Automated purification with the EZ1&2 RNA Tissue Mini Kit requires use of one of the following:

  • the EZ2 Connect
  • the EZ1 Advanced XL and the EZ1 Advanced XL RNA Card

EZ1 cards are preprogrammed with protocols that guide the user through worktable setup and provide the instrument with commands for the purification procedure.

Using silica-coated magnetic-particle technology, the easy-to-use instrument allows purification of 1–24 samples (EZ2 Connect) or 1–14 samples (EZ1 Advanced XL) in a single run. Presealed reagent cartridges and the automatic sensor-controlled lock of the instrument door reduce the chances of environmental contamination or degradation due to RNases. The automated procedure provides high repeatability and linear yields over a range of starting amounts. The flexibility to purify high-quality total RNA from a wide range of tissue types ensures that samples can be purified for a range of gene expression analysis applications. RNA yield using the EZ1&2 RNA Tissue Mini Kit is dependent upon many variables, including sample weight, tissue type and developmental state.

Procedure

Tissues are first lysed and homogenized in Buffer RLT. RNA is then purified in one step on the instrument following the automated EZ1&2 RNA procedure. For certain RNA applications that are sensitive to very small amounts of DNA, an integrated DNase digestion step removes genomic DNA contamination. Total RNA is eluted in 200 µl, 100 µl, or 50 µl, ready to use in all downstream applications.

Is training of lab personnel included with the purchase of a BioRobot EZ1?

Operation of the BioRobot EZ1 is so easy that no additional training other than the initial demo is needed. Operating the instrument requires only three major steps:

  • Insert the appropriate EZ1 Protocol Card, switch on the workstation, and, if necessary, select the appropriate protocol
  • Follow the instructions on the liquid-crystal display (LCD), which describes where the sample tubes, elution tubes, filter tips, and EZ1 Reagent Cartridges must be loaded and which indicates when to start the protocol run
  • Collect purified nucleic acids when the protocol run ends

No, the new EZ1 RNA Kits can only be used with updated protocols programmed on the new EZ1 RNA Card V2.0.

The new card contains all protocols available for the isolation of RNA on the BioRobot EZ1 with the new kits:

Likewise, the former EZ1 RNA Cell Mini Kit (cat. no. 958134), EZ1 RNA Tissue Mini Kit (cat. no. 959134) and EZ1 RNA Universal Tissue Kit (cat. no. 956136) are not compatible with the new EZ1 RNA Card (cat. no. 9015590).

How can we get our BioRobot EZ1 upgraded when new protocols are launched?

Protocols for nucleic acid purification are supplied on pre-programmed EZ1 Protocol Cards. These protocols provide both on-screen instructions for the operator and operating commands for the workstation. Loading of sample tubes, elution tubes, filter-tips, and Reagent Cartridges onto the worktable of the BioRobot EZ1 is intuitive, easy and quick. Details about worktable setup are provided with each new protocol, and are also displayed in the liquid-crystal display (LCD) of the control panel when you start setup of the worktable.

Do the foils sealing the EZ1 Reagent Cartridges have to be manually removed prior to loading the robot?

No, prefilled Reagent Cartridges can be loaded onto the BioRobot EZ1 with their seals. During operation of the instrument, the piercing unit, a row of 6 metal spikes located behind the tip adapters, will puncture the foil of the cartridges, thereby exposing the reagents.

What has to be done to an RNA sample before loading it onto an Agilent Bioanalyzer?

For RNA isolated on the BioRobot EZ1 and BioRobot M48:

The RNA can be directly applied to the Agilent Bioanalyzer, since it is being denatured in the final protocol steps of these isolation procedures.

For RNA prepared with all other QIAGEN RNA Isolation Products:

We recommend to denature the samples in a water bath for 2 min at 70°C, and then place them directly on ice prior to loading them onto the Agilent Bioanalyzer.

How do I safely inactivate biohazardous flow-through material?

Always dispose of potentially biohazardous solutions according to your institution’s waste-disposal guidelines. Although the lysis and binding buffers in QIAamp, DNeasy, and RNeasy kits contain chaotropic agents that can inactivate some biohazardous material, local regulations dictate the proper way to dispose of biohazards. DO NOT add bleach or acidic solutions directly to the sample-preparation waste. Guanidine hydrochloride in the sample-preparation waste can form highly reactive compounds when combined with bleach.
