Product Details

The AllPrep DNA/RNA/miRNA Universal Kit enables simultaneous purification of genomic DNA and total RNA, including miRNA, from cells and all types of tissue, including lipid- and fiber-rich tissue. Since there is no need to divide the sample into two for separate purification procedures, maximum yields of DNA and RNA/miRNA can be achieved. The purified DNA and RNA/miRNA are eluted separately and are ready for use in a variety of downstream applications. The kit includes optimized protocols for various samples types, including tissue and separated blood cells. It can be automated on the QIAcube Connect.

Performance

The AllPrep DNA/RNA/miRNA Universal Kit is the only kit currently on the market for successful purification of high-quality DNA, RNA, and miRNA from the same cell or tissue sample, including difficult-to-lyse fibrous and lipid-rich tissues (see figures " Highest yields of DNA", " Highly pure RNA", and " Reliable miRNA purification"). The kit provides a simple, convenient, and user-friendly procedure that does not require the use of toxic substances such as phenol (see figure " The fast and convenient AllPrep procedure").
High yields of analytes
High yields of nucleic acids can be easily purified from fiber-rich tissues, such as skeletal muscle, heart, and aorta, as well as lipid-rich tissues such as adipose or brain tissue (see figure " Highest yields of DNA"). Genomic DNA purified with the AllPrep DNA/RNA/miRNA Universal Kit procedure has an average length of 15–30 kb, depending on homogenization conditions. In addition, all RNA molecules that are >18 nucleotides can be easily isolated.

Principle

The AllPrep DNA/RNA/miRNA Universal Kit integrates QIAGEN's patented technology for selective binding of double-stranded DNA with well-established RNeasy technology. Specially optimized enzymatic digestion steps enable the purification of RNA, miRNA, and DNA — even from challenging samples.
Biological samples are first lysed and homogenized in a highly denaturing guanidine isothiocyanate-containing buffer, which immediately inactivates DNases and RNases to ensure isolation of intact DNA and RNA. The lysate is then passed through an AllPrep DNA Mini spin column. This column, in combination with the high-salt buffer, allows selective and efficient binding of genomic DNA. On-column Proteinase K digestion in optimized buffer conditions allows purification of high DNA yields from all sample types. The column is then washed and pure, ready-to-use DNA is eluted. Flow-through from the AllPrep DNA Mini spin column is digested by Proteinase K in the presence of ethanol. This optimized digestion, together with the subsequent addition of further ethanol, allows appropriate binding of total RNA, including miRNA, to the RNeasy Mini spin column. DNase I digestion ensures high-yields of DNA-free RNA. Contaminants are efficiently washed away and high-quality RNA is eluted.

Procedure

Preoptimized protocols for various sample types
The AllPrep DNA/RNA/miRNA Universal Kit includes dedicated protocols to ensure optimal yields of DNA, RNA, and miRNA from various starting materials — including cells, tissues, and human blood. This leads to greater efficiency, as there is no need for time-consuming optimization or establishment of user developed protocols and procedures for challenging starting material, such as fibrous or lipid-rich tissue.
Convenient, streamlined procedure — no need for toxic substances
In just 75 minutes, the fast and easy AllPrep procedure delivers high-quality analytes, ready for use in subsequent downstream applications (see figure " The fast and convenient AllPrep procedure"). Due to the innovative kit chemistry, there is no need for toxic substances such as phenol, minimizing environmental and user hazards. The AllPrep DNA/RNA/miRNA Universal Kit can also be automated on the QIAcube, ensuring maximal convenience and standardization.
