Product Details

The Investigator ESSplex SE QS Kit amplifies the full suite of 16 STR markers in the European Standard Set (ESS), including SE33 and the gender-specific Amelogenin, as well as a unique performance control called Quality Sensor (QS). The Quality Sensor provides additional, valuable data for your sample quality control and PCR performance check. By taking advantage of QIAGEN fast-cycling technologies, the kit enables PCR amplification in just 60 minutes. Highly sensitive, with rapid and reliable results from trace DNA, the Investigator ESSplex SE QS Kit is suitable for all forensic applications and paternity testing. The Investigator ESSplex SE QS Kit meets ISO 18385 requirements.

Performance

The Invesigator ESSplex SE QS Kit utilizes fast-cycling technology to provide reliable results in approximately 60 minutes.

The Fast Reaction Mix 2.0 is based on proven QIAGEN multiplex PCR technology that has been further optimized for direct amplification. Assay performance tests in the presence of increasing concentrations of inhibitors showed superior inhibitor resistance. This means reliable, high-quality results.

The Investigator ESSplex SE QS Kit was validated and evaluated using the GeneAmp 9700 thermal cycler (in standard mode) and the Applied Biosystems 3500 Genetic Analyzer.

Principle

The Investigator ESSplex SE QS Kit uses QIAGEN's fast-cycling PCR technology for the simultaneous amplification of 16 STR markers and Amelogenin, as recommended by the European Network of Forensic Science Institutes (ENFSI) and the European DNA Profiling Group (EDNAP) as the European Standard Set of loci (ESS).

The Investigator ESSplex SE QS Kit Primer Mix contains the innovative performance control Quality Sensor (QS1 and QS2). It provides additional information on sample quality and PCR success. The Quality Sensor, being simultaneously amplified with the STR markers, shows you if you are dealing with:

  • Successful PCR amplification
  • Failed PCR amplification
  • Absence of DNA
  • Degraded DNA
  • Inhibitors

The additional information obtained with the Quality Sensor will help you streamline your sample analysis by showing you where to focus your analytical efforts, and save you time and money by eliminating the need for unnecessary PCR re-runs.

Procedure

The 16 STR markers, Amelogenin and the Quality Sensor are amplified simultaneously in a single PCR, and labeled with one of the dyes listed in Table 1 "Investigator ESSplex SE QS Kit dyes and corresponding markers".
The loci then undergo capillary electrophoresis. The results can be analyzed using dedicated software, such as the Applied Biosystems GeneMapper ID-X. See the table "Investigator ESSplex SE QS Kit technical specifications" for more information.
