The PyroMark Q96 ID dispenses the correct volume of enzyme mix, substrate mix, and nucleotides using variable pulse times and dispensing pressures. The settings for these pulse times and dispensing pressures are specific for each lot of PyroMark Q96 Cartridges and correspond to the method number printed on the PyroMark Q96 Cartridge. The instrument method should be selected in the PyroMark Q96 Software or in the PyroMark CpG Software, according to the reagent cartridge that will be used for the run. The settings and instrument method files provided on this page are only for use with PyroMark Q96 ID.
Dispensing pressures and pulse time settings must be checked and, if necessary, updated every time a cartridge from a new lot number is used.
Managing instrument parameters in PyroMark Q96 ID Software
- Click “Instrument” and then “Parameters” to reveal the list of installed instrument parameter files.
- Right-click an existing instrument parameter file from the list and click “Duplicate”.
- Enter a new file name (e.g., “Instrument parameters Code 0005”).
- In the following table, find the dispensing pressures and pulse time settings that correspond to the code number printed on the label of the cartridge to be used.
- Enter the dispensing pressures and pulse times in the corresponding fields of the “Instrument Parameters” dialog box.
- Save the new instrument parameters file.
- When creating a new Run Setup, select this new instrument parameters file (e.g., “Instrument parameters Code 0005”) from the list of instrument parameter files.

Managing instrument parameters in PyroMark CpG Software
- Access the available downloadable instrument method files below or under "Resources" on the PyroMark Q96 ID product page.
- Click the instrument parameter file that corresponds to the code printed on the label of the cartridge to be used.
- Unzip the downloaded file and save the instrument parameter file to the operator computer:
- If using PyroMark CpG SW Version 1.0.9 or lower, save the file to
C:\Program Files\Biotage\Pyro Q-CpG 1.0.X\Instrument Parameters (where "X" represents the installed version of PyroMark CpG Software)
- If using PyroMark CpG SW Version 1.0.11 or higher, save the file to
C:\Program Files\Qiagen\PyroMark CpG Software 1.0.11.14\Instrument parameters
- The downloaded file will now appear in the list of available instrument parameter files and can be selected when creating a new Run Setup.
